Assessment of cellulolytic microorganisms in soils of Nevados Park, Colombia BJM

A systematized survey was conducted to find soil-borne microbes that degrade cellulose in soils from unique ecosystems, such as the Superpáramo, Páramo, and the High Andean Forest in the Nevados National Natural Park (NNNP), Colombia. These high mountain ecosystems represent extreme environments, such as high levels of solar radiation, low atmospheric pressure, and extreme daily changes in temperature. Atividade celulolítica de fungos isolados de bagaço de cana-de-açúcar e madeira em decomposição PAB

O objetivo deste trabalho foi identificar isolados de fungos a partir de bagaço de cana-de-açúcar e madeira em decomposição e avaliar a sua atividade celulolítica em bagaço de cana. Cinco isolados foram avaliados, tendo-se como referências os fungos Trichoderma reesei QM9414 e T. reesei RUT C30. A atividade celulolítica foi estimada pela capacidade hidrolítica do extrato enzimático dos fungos cultivados em bagaço de cana sobre os substratos papel de filtro (atividade celulolítica total) e carboximetilcelulose sódica (atividade da endoglucanase). Biosynthesis, purification and characterization of endoglucanase from a xylanase producing strain Aspergillus niger B03 BJM

An extracellular endoglucanase was isolated from the culture liquid of xylanase producing strain Aspergillus niger B03. The enzyme was purified to a homogenous form, using consecutive ultrafiltration, anion exchange chromatography, and gel filtration. Endoglucanase was a monomer protein with a molecular weight of 26,900 Da determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and 28,800 Da determined by gel filtration. The optimal pH and temperature values for the enzyme action were 3.5 and 65ºC respectively. Endoglucanase was stable at 40ºC, pH 3.0 for 210 min. Activities of cell wall hydrolases of Euphorbia heterophylla L. (wild poinsettia) endosperm were investigated during pre- and post-emergence periods, defined as the time interval before and after 2.2 days from the start of imbibition, respectively. The activities of endo-beta-mannanase and beta-mannosidase are higher over the pre-emergence when compared to the post-emergence period and they may be involved in the process of germination in E. heterophylla. Characteristics of a β-1,4-D endoglucanase from Trichoderma virens wholly applied in a palm-fruit husk-based diet for poultry layers BJM

The characteristics of an endoglucanase produced by a Trichoderma virens strain T9 newly isolated from a palm-fruit husk dump site, its physiological characteristics and enzyme production were studied. Whole cells of the depolymerizing-enzyme producing T. virens were applied to palm-fruit husk and bird performance characteristics when employed as poultry diet additive were considered. Endoglucanase activity in submerged fermentation was 1.6 nkat. Optimum activity was recorded at pH 6.0 and 55ºC. The enzyme retained 50% residual glucanase activity at 70ºC for 10 minutes. 1.0% Tween-80 and SDS yielded endoglucanase activity 2.15 times higher than the control. Activity wasboosted by 20mM Ca2+ (115.0%); 10mM K+ (106.5%); and was totally inhibited by 1mM Hg2+....

Characterization of a thermostable endoglucanase produced by Isoptericola variabilis sp. IDAH9 BJM

Abstract This study aimed to isolate and evaluate the cellulase activity of cellulolytic bacteria in hot springs of Dehloran, Ilam province, Iran. Water and sludge samples were collected from the hot springs and the bacterial enrichment was performed in a medium containing rice barn and carboxymethyl cellulose (CMC). The cultures were incubated at 50 °C in aerobic conditions. The bacteria were isolated on CMC agar (1%) medium. Cellulase assay of the isolates was measured by the evaluation of endoglucanase enzyme activity, which is also called as carboxymethyl cellulase (CMCase). Cellulase is a group of enzymes (endoglucanase, exoglucanase and beta-glucosidase) required for cellulosic feedstock hydrolysis during bioethanol production. The use of recombinant cellulase is a strategy to reduce the enzyme cost. In this context, the present work describes the construction of a cellulase expression vector (pEglABglA), which allowed constitutive co-expression of endoglucanase A (EglA) from an endophytic Bacillus pumilus and the hyperthermophilic β-glucosidase A (BglA) from Fervidobacterium sp. in Escherichia coli. Multivariable parameter optimization for the endoglucanase production by Trichoderma reesei Rut C30 from Ocimum gratissimum seed BABT

The aim of this study was to evaluate the interaction effects of the physico-chemical parameters on the endoglucanase (CMCase) production by Trichoderma reesei Rut C30 on a cellulosic agro-residue by the solid-state fermentation (SSF) and to determine their optimum values by the EVOP factorial design technique. The best combination of physical parameters for the maximum production of the endoglucanase (CMCase) was 28ºC temperature, 79% relative humidity and 4.8 pH of the medium. The best combination of the chemical parameters was (mg/L) nicotinic acid 15, naphthalene acetic acid 7, ferric chloride 5 and Tween-80 6. With the application of this technique, the yield of the CMCase increased by ~ 2.3 fold.

Cellulase is a complex enzyme system, commercially produced by filamentous fungi under solid-state and submerged cultivation. It has wide applicability in textile, food and beverage industry for effective saccharification process. In this study, cellulolytic enzyme activity, particularly endoglucanase of 26 Streptomyces strains isolated from garden soil was examined, including two isolates selected on the basis of potential cellulolytic activity on Bennett's agar medium. Study on Thermodynamics and Adsorption kinetics of Purified endoglucanase (CMCase) from Penicillium notatum NCIM NO-923 produced under mixed solid-state fermentation of waste cabbage and Bagasse BJM

In the current study, one thermostable endoglucanase was purified from Penicillium notatum NCIM NO-923 through mixed solid state fermentation of waste cabbage and bagasse. The molecular weight of the purified enzyme was 55kDa as determined by SDS polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme had low activation energy (Ea) of 36.39KJ mol-1 for carboxymethyl cellulose hydrolysis and the enthalpy and entropy for irreversible inactivation was 87 kJ mol −1 and 59.3 J mol −1 K−1 respectively. The enzyme was quite thermostable with a Tm value of 62.2˚C. The pKa1 and pKa2 of ionizable groups of the active sites were 2.5 and 5.3 respectively. Apparent Km, Vmax and Kcat of the enzyme were found to be 5.2 mg mL-1, 80 U/gds and 322.4 sec-1 respectively....

Variáveis que influenciam a produção de celulases e xilanase por espécies de Aspergillus PAB

O objetivo deste trabalho foi determinar as variáveis que influenciam a produção simultânea de celulases e xilanase por Aspergillus aculeatus URM 4953 e A. phoenicis URM 4924, com uso de bagaço de cana-de-açúcar como substrato. As variáveis avaliadas foram pH inicial, tempo de cultivo, concentração do substrato, agitação, concentração do inóculo, temperatura, tipo de bagaço de cana-de-açúcar e espécie de Aspergillus. Foi utilizado o planejamento fatorial fracionário 2(8-4), com três repetições no ponto central. Todas as variáveis analisadas influenciaram a produção de pelo menos uma das enzimas.
